Abstract

The objective of this chapter is to introduce the polarization microscopy and focus on its clinical applications. The polarization microscopy works on a principle that when two orthogonally polarized light rays pass through orderly arranged filamentous structures, they exhibit birefringence. Polarized microscope system has been designed to observe an optically anisotropic specimen. This specialized microscope has been developed as a variant adjustment of the standard light microscope equipped with a polarizer, motorized micromanipulator, and an analyzer. Polarized light microscopy enhances the contrast between the cell and the background, which then helps to reveal specific important information about the structure and the composition of the substance. This chapter covers the components of polarization microscope and its functions, differences between the simple and polarizing microscope, and advantages and disadvantages of the polarizing microscope.
