Abstract

Replication is defined as the process by which copying of base sequence present in the parent strand to daughter strand, thereby passing the genetic information from parent to progeny. DNA replication is the biological process of producing two identical replicas of DNA from one original DNA molecule. This process occurs in all living organisms and is the basis for biological inheritance. This chapter discusses enzymes involved in the DNA replication, DNA polymerases, steps of DNA replication, and DNA repair mechanisms. Enzymes involved in the DNA replication are topoisomerases, helicase, single strand binding (SSB), DNA primase, DNA polymerase, and DNA ligase. Three important properties of DNA polymerase complex are chain elongation, processivity, and proofreading. The mechanisms of DNA repair include nucleotide excision repair (NER), mismatch repair (MMR), base excision repair (BER), homologous recombination (HR), and nonhomologous end-joining (NHEJ) repair.
