Abstract

The aim of this chapter is to focus on the estimation of serum bilirubin. Bilirubin is the break down product derived from the heme part of heme compounds like Hb, Mb, cytochromes, catalase, peroxidase, etc. Heme is catabolized in reticuloendothelial system, mainly in spleen and liver. This bilirubin is unconjugated and is insoluble in water. It is transported in blood bound to serum albumin and is taken up by hepatocytes through a carrier mediated process. Bilirubin reacts with diazo reagent to form a colored complex which is measured at 540 nm photometrically. Conjugated bilirubin (direct bilirubin) undergoes color reaction directly, whereas unconjugated bilirubin (indirect bilirubin) forms color after addition of alcohol. Total bilirubin is the sum of direct and indirect bilirubin.
