Stool examination is done to find out ova and cysts of different parasites. Saline preparation as well as iodine preparation of stool is made on the same glass slide. Each preparation (saline and iodine) should be covered with a square coverslip, taking care that no air bubble forms. It is easier to identify eggs of cestodes, helminths and protozoa. Therefore, always prefer to search for these abnormal findings. Before adding feces either to saline or iodine drops, feces in container should be mixed properly with wooden stick. If preparation has dried up, prepare another slide for examination purposes. A detailed discussion on identification of eggs and cysts microscopically has been presented in this chapter.
