The embedded tissues are then subjected to section cutting. This involves the technique of microtomy which yields thin sections of the tissue for microscopic examination of the internal structure. Sections are prepared on an instrument known as a microtome of which various types are available. Common features of all the microtomes are that they are having firm support for knife and tissue block and a feed mechanism designed to advance the specimen a predetermined number of micrometers on each cutting stroke. This chapter covers the microtomes, microtome knives, section cutting, attaching the sections to slides, and problems and remedies encountered during mounting of the sections.
